Re-rolling is simply restarting the game from the point where it would generally be like a freshly installed game without any records of progress. The point of doing this is to somehow manage to earn the best set of Summon results from the initial free draws that you'll be given when starting the game. In this particular game, you'll easily get a huge chunk of the premium currency called Wyrmites from the get-go which will allow you to draw at least (2) ten-fold summons right from the start (by jumpstarting on Endeavors, you'll earn 325 wyrmites, plus the 2800 you'll get from the giftbox from the start, you'll have a total 3125 wyrmites).

Why do I need to re-roll?

Adventurers, Dragons, and Wyrmprints in the game are the units you basically use to play through the levels of the game. Each one is designed differently and are mainly classified into two important things: by their elemental affinity, and by rarity. Needless to say, the better rarity the unit has, the better starting base stats it also has -- which means it will generally benefit you best to have them in your roster.

Re-rolling can quite be tedious as clearing the cache of the installed game would generally mean you'll have to download the in-game contents all over again -- at least for this game (which is why you'll need a wi-fi connection to do it). However, it is to your great advantage to have at least a 5-star adventurer when starting the game, as it is relatively difficult to collect more of them later on in the game, as well as the Wyrmite needed to actually Summon them.

What you shouldn't do when re-rolling

The thing that you might want to avoid most is linking your Nintendo account prior to getting the 5-star Characters that you like. Doing this would save your progress in their server -- which would make it impossible to perform re-rolling any further, unless otherwise you'll be using a different phone/account.

Additional Tips:

When pulling out your draws in the Summon section, a 5-star pull will show a rainbow colored gear on its animation. To speed up on the process, you may also skip the entire animation phase to instantly check on the stuff that you earned from the summon. If you failed to get a 5 star summon on the first ten-fold pull, try the second one prior to re-rolling the game. You can't be greedy trying to get more than two 5-star pulls or you'll find yourself doing this over and over again.
