No the multiplayer only works when you are connected to Xbox live. At least that is the only way I can get on it and when I read the little booklet that came with the game (yes I read it) that is what I understood. So, in short, it is suppoed to do that.

The start of this mission is all about stealth, with the many MV's around, you have to get as far as you can withpout being caught, then when you are seen, get into one of the MV's by the side, and don't bother fighting too much, just get through the level.
