Well, if you haven't rescued all the Little Sisters, I suggest you do that for the achievement. It doesn't matter what you do, but if you harvest them, you get more Adam. If you rescue them, you get some Adam, but if you rescue three or four in a row, Tenebaulm will send a teddy bear to a Gatherer's vending machine, which will have good stuff in it. She will send you Plasmids that you won't be able to find, tonics, more Adam, money, ammo, and other good stuff. If you harvest, you get the bad ending, but it's still good to see.

Do you mean fort frolic? And if you do then you need to put the concation in the machine before being able to open the door to the next bathasphere.You remeber the room you were in to get the recording of how to creat the latheras specter right?;but if that isnt it try to check the radio messages sent to you maybe you forgot something,or maybe the game glitched so you have to restart from the last save.

If you spliced the Fire plasmid then it would still be in your actual inventory even if you switched it out. All you have to do in that case is access a gene bank and you can re-equip your Fire plasmid and anything else that you want.

I guess if you would have sold it, I'm pretty sure you can't either way, then yes, you would have to go to the Gatherer's Garden and use more Adam to buy the new plasmid.

Um...no. You can either have the game auto save which is when passing from one section of Rapture to the other, or manually saving in your pause menu.
