When you clear the indicated mission in Single-Player the corresponding boss character will become an available option as a playable character.

Unlock Biker Ben:

Beat Biker Ben on the Training level 6.

Unlock Snorkel Jane:

Beat Snorkel Jane on mission 8 of the Beach.

Unlock Trooper Tim:

Beat Trooper Tim on mission 8 of the Plaza.

Unlock Cavegirl Carmen:

Beat Cavegirl Carmen on mission 8 of the Nature Park.

Unlock Rabid Rabbit:

Beat Rabid Rabbit on mission 8 of the Playground.
