Pikachu and the Pokemon Trainer are available from the start.

Lucario can be unlocked in three different ways. Have Lucario join your team in the Subspace Emissary, play 100 matches in Brawl, or beat the Target Test for one character on all difficulties. With the exception of the Subspace Emissary way, you must defeat Lucario in a fight before you will unlock it.

Jigglypuff can be unlocked in three different ways. Play 350 matches in Brawl, beat the Subspace Emissary and then return to the correct area (I don't remember which), or beat the Subspace Emissary and Events 1-20. You will need to battle Jigglypuff in a battle before you can unlock it.

Pikachu and Pokemon Trainer are default characters. For Lucario complete The Glacial Peak stage in the Subspace Emissary or play 100 matches or Complete Target Smash on all five difficulties. For Jigglypuff, go back to the Swamp after you completed the game in the Subspace Emissary and look for a door in the second half of the stage, or play 300 matches, or complete 20 events after completing the Subspace Emissary.
