no you cannot,it's a one time deal for each trial

Yes, you can restart a Cloister of Trials. The only one you can't is the one in Bevelle because you're not allowed back in there after you leave. All the other ones though, you can go through again. At the end of the Trials, where the Aeons sleep, are a few treasure chests.

You CAN repeat the cloister of trials as many times as you wish (Aside from the one in Bevelle, because you can't go back there). When you repeat them (after you gain the airship), there are multiple treasure chests in each Faith Room containing alleged "Gifts" from all of the summoners that have visited the temples.
