Do you mean unlock it as in being able to switch between it and regular costume? Or just get it in game? Either way, halfway through the storyline Peter Parker goes back to his apartment, and his suit turns into the black spidey suit. You haven't unlocked it yet, Spiderman just wears it for a while. After you beat the game then you unlock the Black suit, then you can play through the whole game in the black suit, if you so choose to do so.

For this you will have to complete 2 missions and have to climb 50 buildings. When you will climb 50'th building you will automatically get athe black suit
