You have to win all the other characters before you can get Mr. Game and Watch.

Just beat all the target practices. The last player you beat them as will be the one you fight him with.

There is no fastest way; to get Mr. Game & Watch, you have to do one of the following:
- Complete Classic Mode with all of the other 24 Characters
- Complete Adventure Mode with all of the other 24 Characters
- Play 1,000 Melee vs. Matches
