To get Mewtwo, you have to play a combined total of 20 hours in multiplayer mode. Also if you have 2 controllers in, it's reduced to 10 hours. So you could just leave it on all night with 2 controllers in, and then have the winner fight(and hopefully unlock) Mewtwo.

Play vs. Mode for 20 hours or more, combined by all the controllers plugged into the GameCube. For example, if you start a battle with 4 controllers connected to the characters on the field and you leave the game alone for 5 hours, that will be 4 controllers x 5 hours which equals 20 hours.
Immediately after the battle ends, Mewtwo will challenge you.

Yes just go to vs mode leave it there and come back when 20 hours are up
