Be sure to search everywhere thoroughly, you will find that even areas that appear empty will have items to find, enemies to kill and boxes to break. Pay attention to the glowing spots you see scattered around your map as these indicate hidden items. You will generally find the chests in hard to reach areas.
Make getting the 'Gum Gum UFO' skill in the 'Explore' section the first skill in the Skill Tree you grab as it will enable you to easily reach higher areas that would otherwise be difficult to get to.
Save yourself a lot of time by using fast travel whenever possible to reach certain parts of the map in seconds. There is no limit to the amount of times you can fast travel and you will not be penalised.
Oddly, Luffy who is a pirate can't swim. Make sure he doesn't fall into water otherwise you will find your screen fading to black and being replaced just before you entered the water.
