First off, you need access to both sides of the island to get a 6 star wanted level.
1) Activate a Rampage mission, and give headshots to anybody nearby, without completing the mission. Then wait for the Rampage timer to reach zero, and pick it up again. This is easier if you're in an unreachable/semi-unreachable position, like the rooftop of a building.
2) If you've collected all 100 Hidden Packages, you can use the Apache-like chopper in the army base (requires you to wear the Cop uniform). Just go around shooting your missiles at anything that moves.
3) Rob a store until your wanted level reaches 6 stars.

I stood in the stadium. On the second island. Then I shot everything that came my way. And as the tanks, army trucks, cop cars drove around down the bottom they flipped caught on fire and blew up causing my wanted level to be six.
