Yes, 2 people can co-op play Star Trek offline on the Xbox 360 console. When the screen directs you to "Choose Your Character" between "Spock" on the left and "Kirk" on the right, in the bottom left corner of the screen it says "Player 2 Press (START) To Join"; Player 2 should be signed in under a different profile (as there are achievements for you both.
