I am on the mission where I am supposed to me the reporter guy at the Marina/Mikes bar. This is after destroying the pipeline. I have the red mission ring over the Marina bar, but when I show up, I can't enter the bar from either the front or the back door. Is this a glitch?

Yes this could be a glitch or it could be a matter of showing up at specific time, try coming back later. If this does not work check your disk out it may be scratched somewhere quite deep which can cause problems so if that is the case go get it fixed or clean it.
