You just stand within the markers you are shown. You may need to enter Eagle eye/vision to see them. I cannot remember, it's been a while since I've played the game.

Look at the ground in front of the stage, you will see three small patches of hay lying there. Go stand on the one that is still free after the other actors (two groups) appear in front of the stage. At the cue walk clockwise to the next patch, and after the second cue walk clockwise to the last patch. You should now be standing on the patch directly in front of the stage and I believe a cutscene appears. Use your Eagle Vision to determine which soldier you're going to have to kill. Wait for the last cue to walk slowly onto the stage and kill your target.

Addition to the last post - you can ONLY kill your target with the hidden blade and ONLY from behind.

Use the eagle vision and then there will be Brightblue/ white patches. You stand on these until they dissapear then move to the next and the next. Once the timer pops up kill the person that appears gold in eagle vision
