You will not be able to kill Lady Dimitrescu to begin with, your bullets will only stagger her momentarily. It will not be until you progress the story enough to where she transforms, and a boss fight ensues that you can permanently take her down. Trying to kill Lady Dimitrescu before the story permits you will simply result in you wasting ammunition, she does not drop any items when staggered.
