I don't know exactly how a game doesn't allow you to save, it should save whenever you complete a mission or sleep. However it is possible that you activated cheats, you can't save when cheats are activated.

For one, make sure cheat codes aren't active. If cheats aren't on, you should be able to save. But alas, you still wont be able to save. Sometimes the controlled will "shut off" certain buttons. The best way to reset it is to turn it off and then turn it back on. If it still doesn't work, then you will need to wait a little bit until the button resets itself. If that doesn't work, then you need to reload your game. Those are some of the best ways to get the save icon to appear.
