I want just wanted to know if this trophy can be obtained on the first walkthrough, or it can be done in a new game + mode, with all the upgrade and weapons.

Yes it can be done in the first play through but you'd have to be a freaking genius to figure out all the enemies and puzzles quickly... And sadly no the upgrades can't be carried into a new game file:(
