I have all of the characters unlocked but I haven't unlocked some guys who were in the story. I wanted to know if you could unlock the leader of the Ghostown, Jesse Lynch, or Holstein Hals brother, Longhorn Luke. Please tell me if you can.

I'm not 100% sure but I dont think you can unlock those charactors.
You can only unlock the leader of ghost town which is Mr. Black.
You can unlock mr.black and longhornluke and I am 95% sure that you can unlock jessie lynch!!
No, you cannot unlock those characters. I am sure that you have not unlocked the last character Manny Quinn he is the last character.
