Your companions do take damage and when their HP is depleted, they'll be knocked down. You can still revive them by reaching them before the timer expires. If you can't they'll be out of commission, and you won't be able to call for them until you revive them from the Roster menu. Reviving fallen comrades will require medicinal herbs. These herbs can be acquired in the wild or harvesting them from the Healing Garden in your homebase. This facility in your homebase can have more herbs by upgrading it and the herbs respawn regularly by fast-travelling to your homebase.
