All other portals have a glowing effect but the third portal that allows you to go threw the last portal to last island is not glowing so I can not activate it to use. Help please I'm at a stand still and do not want ant to start game all over.

Continue completing quests. I can't remember exactly which ones, maybe with the newlyweds. All of a sudden it's active and you have a new set of quests for that section.
