That is a hint. To open the door to Queen Ghoma you must stun three Deku Scrubs. This is the order to beat them in.

I think it could also be a reference to Micheal Jordan haha I mean, seeing as how many games contain pop culture references.

2 3 1, it means kill the middle second deku scrub the the right third one, then the left first one... 2, 3, 1.

It's a pun in the original Japanese, badly translated. The deku scrub says ニイさんいちばん, which sounds like "2 3 1st" but really says something like "Big Brother is the best."
