On the plane there is a gun pick up, use that to enter a cutscenes, follow the instructions to the bottom of the plane. Go to the red box as soon as you can once the creature comes back.
After the plane? If you mean Ustanak (the Nemesis type thing) you can either use up all your ammo trying to defeat him or you can climb on the cargo crates, push the containers of pressurised containers off and shoot them when he's near them.
