To get a one shot kill on a Witch you have to be using a shotgun, and approach the Witch slowly, just so she almost standing when you're in front of her when she's just about up, blow off her face.

As for the Tank, your team mates are always going to shoot at it, so if you're online with friends, ask them not to, if offline you'll have to kill your AI controlled partners in the safe room. It'll be easy if you set it on fire, and give a bit of bile.
