I want to know what do you do if there no one in big town and you didnt kill them?
the other questiom is do you have to do trouble on the homefront when you're doing the main quest or can you do it after you beat the game

If theres nobody left in big town that probaly means the super mutants killed them all and you can do trouble on the homefront any time
If there is no one there, they are all dead, and there is nothing for you to do there now. As for trouble onthe homefront, you would need to do it before you beat the game, unless you have the dlc broken steel, or you have the game of the year addition,(you can tell if you look at the cover of the bax), which comes preloaded with all of the dlc(s)(theres more than 1).
1. Normally, if you leave the people of Big Town without training them, the Super Mutants will kill everyone there. Sometimes not, but usually. Even if you train them/help repell Super Mutants, they still might just disappear, sorry.
2. As far as I know, you can do "Trouble on the Homefront" at any time after completing the quest "The Waters of Life".
With trouble on the homefront if you are having problems picking up the vault 101 broadcast could be the exp. Packs causing a problem, if you go to the Pitt then straight to vault 101 should be able to start the quest, well worked for me
