You need to complete all the lessons (in all 4 years) go back to the room blast it. The trap door will open. Make sure you have a dark wizard with you (snape will work use the pollyjuice once you have bought him). The trap door has a bad habit of glitching shut so make sure you only go in once you have everything you need.

There are actually two doors into the Charms classroom. If you enter the classroom through the right door, the question mark trapdoor will only open with a key character. If you enter through the left side, you need to use Reducto to blast the lock open, and once you do this and it has been blasted through, it will slide open when you return from this left side.
