You have to get the Mystic Melody for each character, it's just like any other upgrade. Go to:

The mystic melody is a permanent upgrade, and an essentual one, that is used to activate an Ancient ruin. Ancient ruins are those small, alter-looking things with those green gems on each side. When activated through the Mystic melody, well, there's no telling what can happen, unless you've already found out.

This is a very powerful permanent powerup. It is considered so powerful because you cannot, and will not be able to finish all of the missions of the game without it. Using it is one thing, but finding it is something else... There is one for each character, so find them all.

Also, in case you didnt' know, there is a permanent powerup in every level but not mission, so if you go through a level and haven't found a powerup, then you know that there is one there that you haven't collected. Scour the levels.
Be aware though that three levels do not contain powerups: Route 280, Route 101, and the Bonus stage "Green Hill."
