It's not a one time only quest and yes you can repeat the quest every New game you start.

How do you get to deo pass I can't find it anywhere

Deo Pass is located on Rugnica , northeast from the Kaitzur Naval Port , and leading to Akzeriuth
If you are unsure of a certain Area's Location, either:
1. Check your World Map,
Or
2. Check your "Synopsis" under: LIBRARY in the Main Menu.
And it should tell you where to look.
This doesn't work for every area, but it helps with most of them.

How do you find anise after the chat with the elder

Go out of st.binah then head southwest till you reach a bunch of rocks thats where you want to go.
