I remember running across the same problem, you need to talk to Umbracano, I think you already need to have collected a certain amount of the Ayleid artifacts, and then he gives you a quest and you have to meet him at Nenalata, you take him to Nenalata Sel aran arpena and he opens the secret door,
I went into Nenalata because I read a book on a shelf in the Arcane University which mentioned that Nenalata was believed to be the burial place of the last Ayleid king.
I thought, "ooo, sounds exciting!"
I should've known it was too good to not be part of a quest I hadn't started yet!!
