The difference(s) between Fable and Fable: The Lost Chapters is that Fable: The Lost Chapters has exactly two hours more storyline than the regular Fable, and also has more map for you to explore.
If you had to choose between the two, get Fable: The Lost Chapters, but, igf you already have Fable, don't bother with Fable: The Lost Chapters

Also, there's an addition of a whore house, new weapons, armor, silver keys, silver key chests, enemies, and demon doors. There are a few new side quests, but nothing much. Just a really annoying quest to save the son of the chief of Knothole Glade. You can also now choose to frame Lady Gray for the Murder of her sister, there's a new hero doll, and lets not forget the new expressions, including the Air Guitar.
If you have Fable, trade it in for Fable: The Lost Chapters. The new content isn't much, but it sure is fun. And when are you going to get another chance to kill a snow balverine?
