First, you do the song, and then push change instruments
Your mii/miis should be there
And if you have done the song in the past-- you just put the pointer over the one you want to change and push B

simmaster answered:

When doing a custom jam - you can play as yourself on all of the instruments, or another mii if you have the data of you playing that song saved (eg. When you play a song as one mii in single player, you can play as another mii and use a mii from a previous time when you played).

You can choose any miis you, or others around you, wish to play as in multiplayer also.
