You can either play 400 brawls, then Toon Link will challenge you. If you beat him you unlock him.
Or Complete the Subspace Emissary, Then play Classic mode with any LoZ character (Link, Zelda, Ganon) and Toon Link will challenge you. Beat him, and you unlock him.
There's some third way you can do if you completed the Subspace Emissary. I think you go back to the Forest stage and there's going to be a door that wasn't there before. Go in and beat Toon Link, and you unlock him

Go to the forest level after beating adventure mode and then you should see a new door that will lead you to toon link

After completing subspace emmiscary go to the forest level you will see a door that you havnt saw the first time you went on that level
