I answered this for someone else a long time ago (it's lost in earlier questions) So here it is -

Costington can be found standing in front of his mansion at the top of Keystone Drive. If you aren't familiar with how to find Keystone Drive, here it is in detail.

Town Square can be found past the jail to your left and you will see a well in the middle of the road (go to the left of the screen) You will also see the Mystery Machine parked in the Square (at the bottom of the screen) Once you are standing at the well and head to the buildings in the upper left corner (there is a table with lit candles on it in front of the staircase.
- Go up the stairs, onto a roof, use the switch on the wall (as Velma of course)to unlock and open a gate, (it will say "The Detour" on the screen as the gate opens) (Press the "C" button on the nunchuk to change to Velma)
- Cross a wooden bridge, a roof and go down a ramp,
- Cross the bit of road to a staircase, take it to a roof (you will encounter a jumping Sombereo with a frog underneath - stop him using the "Z" button he will have interesting things to say - nothing useful but stupid jokes)
- Jump off the roof onto an awning and to the ground (or just to the ground).
- Head down the street (top of your screen) towards the 2 big gates. You now need Daphne, there is a pink pipe on the right near the corner of gate going up to a small awning with a gear on it;
- As Daphne, climb the pipe ("Z" button) and retrieve a gear.
- Jump off the awing with the gear and simply toss the gear at the switch when you are right in front of it
(You only need to do this once - after the first time you just use the switch to get through the gate.)
- Open the gate by pulling ("Z") the switch
- Go through the open gate.
- You now need to be Fred,
- Go over to the box (Anything important you need to find will have what looks sort of like 'bubbles' coming up around it to help you find it.) under the portico and pull it out, jump on it to the awning and then to the roof.
- Cross the wooden bridge, cross the roof, go down the stairs and you have reached "Keystone Drive"
- Go straight ahead to Costington's Mansion (watch out for the bomb shooting pinatas, Mariachi Skeletons and Zombie Brides).
- Climb the stairs onto the Costington Compound, you will find a closed gate and an intercom on the right side of the gate.
- Go to the intercom and press "Z" - Costington will come on and there will be some conversation before he will let you in (once again the gate will be open from now on)
- Continue up another staircase, around the empty pool and you will see Costington standing at the front door (that's where he will be throughout the part of the game in El Muncho) Hope this helps - Have fun!
