To unlock kisame I had to beat single player with Might Guy. To unlock Orochimaru and Itachi I beat single player with kakashi and I unlocked both of them.

Well...
I unlocked Kisame when I beat Single Player with Might Guy.
To unlock Orochimaru, I beat Single Player with Shikamaru, (I also got Gaara and Temari)
My game might be different from yours, who knows? I haven't figured out hot to unlock Itachi yet, but I think you have to complete all missions, genin, chunin and jonin. (yup, a real pain)
