This can be accomplished by either completing Story mode or by playing every board except DK Jungle Ruins in Party mode.

Complete the following tasks to unlock the corresponding effect.
Unlock Kamek:
Beat Story mode.
Unlock Shy Guy:
Beat Story mode.
Unlock Bowser Station Board:
Beat Story mode.
Unlock DK Jungle Ruins:
Unlock for 500 points.
Once you beat story mode three times with koopa, by the koopa constellation and highlight it. Then click on it and paratroopa will fly out or spiny blue koopa will fly out and joins the party.
*note* paratroopa appears if you do koopa. Spiny blue shell koopa appears if you do it 5 times and you can only get one of them so pick wisley.
Ok to do this all you have to do is shake the Wii Remote before hitting the dice block on your turn.
NOTE:
This works on any dice block. And if I were you, I wouldn't do this on the slow dice block because it will make the numbers go by faster and it doesn't stay big so you'll have to keep doing it if you want to keep it big.
