When you complete ALL the levels on the indicated difficulty setting it's respective Time Trial level will become an available option.

Unlock Treasure Room:
On the Easy difficulty setting complete every level on Grad Student.

Unlock Bottomless Pit:
On the Normal difficulty setting complete every level on Archaeologist.

Unlock Harem:
On the Hard difficulty setting complete every level on Adventurer.
