Why do building randomly have no labor when I have enough people and they r happy

It's probably your criminals. If you look closely at the circle above the building you'll see there's a little noose in it instead of just a line like when you don't have enough labor. Build a courthouse, a few guard posts, and a punishment of your choosing (humilation mask, stocks, etc.) Then your criminals will be caught and rehabilitated (speed of rehab depends on punishment) and once they are rehabilitated the place will be up and running again
