I believe you get it by luck, it doubles th EV points you eaar for defeating a wild pokemon.

You get infected by Pokerus whenever the game wants you to be. In other words, you need to have luck on your side. When you do get pokerus, the Pokecenter nurse will tell you that your Pokemon is infected with it. While you are infected with pokerus, your Pokemon will level up twice as fast, and whenever he grows a level, his stats will go up higher than they normally would.

Actually it is really easy to get in ruby and sapphire so transfer a pokerused Pokemon from there to diamond. If not, get lucky!
