No, unfortunately, you cannot. The only Pokemon games where you can unlock a new region is the Gold, Silver, and Crystal series, where you could journey to Kanto after travelling through Johto.

No, there is only Sinnoh in Diamond/Pearl. However, once you beat the Elite Four and obtain the National Dex, you CAN visit the Eastern Island, whose terrain is MUCH different from Sinnoh. But I wouldn't consider it a region.
