Weight determines how much you or the other person will be pushed when you collide. It also determines how much you will be slowed by grass or other things like that.

AND1 answered:

There are two main points of weight:
1. When bumping onto other karts, the weight determines the amount of recoil taken back, ex. Bowser would have less recoil or less feedback.

2. When using the mini-turbo boost, the weight determines how long your boost will be ex. A low weight character and car like Dry Bones in the Banisher or Egg1 will have a long boost because of their low weight.
