In order to rescue some of the harvest sprites of the watering team you have to water a certain spot of land. So my question is does that spot of land need to be tilled?

Yes, the squares must be tilled with your hoe in order to find them. Luckily, they aren't in your farmland but unluckily a confirmed place for where they are is in Galen's massive yard.
Ummm... OF COURSE it needs to be tilled!!! How can you water an untilled land???
