Destroy the vines keeping the tree up with the Boomerang to gain access to the basements. Follow the path from the bottom floor to find a circular room with 4 switches in it. Use the Boomerang to activate the 4 switches to gain access to the big key, but then 2 Moblins appear who you'll have to kill. Kill them and collect the key.

You won't get the Big Key until near the end of the Forbidden Woods Dungeon - you can't miss getting it. Once you use your Boomerang on the 4 vines to knock down the flower that you use float down the river using your Deku Leaf, then you'll be close to getting the Boss Key!
