You don't unlock it, but to play it, go to the 1-Player mode, then go to the mode that has Target Practice(I forgot the title of this mode, sorry). There should be some selection about melees, so go to that, and go to the Cruel Melee and select it.

You don't need to unlock it. During Cruel Melee, you are fighting the toughest opponents in the game without any items.
