You have to do Adventure Mode on Hard or Very Hard - I suggest Hard!!!!!

Actually you can do normal adventure mode with any amount of lives and I think you have to beat it in under 20 minutes.

Do Adventure Mode without using any continues on Normal or above in less than 18 minutes and Bowser at the end will come back after being defeated as Giga Bowser!

The way you can get Giga Bowser in adventure mode is in normal, hard and very hard. Just be sure to be at final destination with three lives or more, he might not come out if you have less.
