To keep M. Valentine you have to have no harpies feather duster or harpie lady in your deck.Get rid of them to unlock her again

To add on to what kool said, once you beat her, you are able to have Harpie cards in your deck and still be able to fight her. As long as you beat someone you unlock, you do not need the requirements to fight them again that you needed to unlock them in the first place.
