Collect the dungeon card in Mission 5 and then complete the mission. Then when you go to the 'Select' screen the extra mission called 'Dungeon' will be available to the top left.

You need to go all up in every door and shoot the barrel in the final mission (aka Mission 5)
It will give you the Dungeon Card then the mission will be unlocked
