You can get the mission item Eldas Cup by doing the dispatch mission that requires Caravan Musk. Just Keep Checking the pub mission board.
To defeat the Queens second form (the Li-Grim) your best bet is to keep attacking with powerful moves and combos. Dont waste your time on the totema, as it is possible to win before they cause to much trouble. Try to avoid using things like ultima as they don't affect the Li-Grim.
To answer your second question; combos. Combos are the key to beating Li-Grim. The Idea is too get Marche and other powerful characters, surround her and pull off three way combos and the like that can deal damage over 300.
